Atopy affects LPS responsiveness and TLR-4 expression in children peripheral mononuclear cells

چکیده
Methods Over 350 subjects were recruited from the Pediatric Test Center of the Montreal Children’s Hospital and a questionnaire was administered to determine presence of a history of atopic diseases. 3-5cc of anti-coagulated blood was taken and peripheral blood mononuclear cells were cultured for up to 24h with IL-4 (13.5ng/ml) and/ or LPS (up to 5μg/ml). We conducted flow cytometric analysis for surface TLR-4 expression, along with T ‘helper’ lymphocyte marker CD4, pan-B lymphocyte marker CD19, monocyte marker CD14, and for intracellular phosphorylated p44/p42 and p38 signaling molecules.

Toll Like Receptor 2 and 4 Expression in Peripheral Blood Mononuclear Cells of Multiple Sclerosis Patients
Background: Multiple sclerosis (MS) is a T cell mediated autoimmune disease with unknown etiology. Appropriate MS therapeutic strategies need thorough understanding of both disease etiology and pathogenesis mechanisms. Ligation of TLR-2 and TLR-4 stimulates the production of several cytokines leading to CNS autoimmunity and neurodegenerative diseases. Objective: To find a relationship between M...
متن کاملIncreased Responsiveness of Peripheral Blood Mononuclear Cells to In Vitro TLR 2, 4 and 7 Ligand Stimulation in Chronic Pain Patients
Glial activation via Toll-like receptor (TLR) signaling has been shown in animals to play an important role in the initiation and establishment of chronic pain. However, our ability to assess this central immune reactivity in clinical pain populations is currently lacking. Peripheral blood mononuclear cells (PBMCs) are an accessible source of TLR expressing cells that may mirror similarities in...
متن کاملIncreased Responsiveness to Toll-Like Receptor 4 Stimulation in Peripheral Blood Mononuclear Cells from Patients with Recent Onset Rheumatoid Arthritis
BACKGROUND Cell signaling via Toll-like receptors (TLRs) leads to synovial inflammation in rheumatoid arthritis (RA). We aimed to assess effects of TLR2 and TLR4 stimulation on proinflammatory cytokine production by peripheral blood mononuclear cells (PBMCs) from patients with recent-onset RA, osteoarthrosis (OA), and healthy control (HC). METHODS PBMCs were stimulated with LPS, biglycan and ...
متن کاملIncreased Expression of Toll-Like Receptors 2 and 4 in Renal Transplant Recipients that Develop Allograft Dysfunction: A Cohort Study
Background: The incidence of ischemic reperfusion injury (IRI) in early phase post-transplantation and activation of toll-like receptor (TLR-2) and TLR-4 remarkably impact the outcome of a renal allograft. Objective: To investigate whether the expression of TLRs in peripheral blood mononuclear cells (PBMCs) can predict the clinical outcome of kidney allografts. Methods: We obtained blood sample...
متن کاملEffect of A-769662, a direct AMPK activator, on Tlr-4 expression and activity in mice heart tissue
Objective(s): TLR-4 activates a number of inflammatory signaling pathways. Also, AMPK could be involved in anti-inflammatory signaling. The aim of this study was to identify whether stimulation of AMPK could inhibit LPS-induced Tlr-4 gene expression in mice hearts. Materials and methods: Heart AMPK activity and/or Tlr-4 expression was stimulated in different mice groups, using respectively IP i...
